The STEDYCON can be controlled via Abberior Instruments STEDYCON smart control, a web-browser-based graphical user interface. STEDYCON smart control was designed from scratch to provide the highest possible usability completing the overall STEDYCON concept of easy installation, intuitive usage, and maintenance-free operation. Beginners can image with super-resolution within minutes. Smart control runs on every device with a modern web browser like Windows-based PCs, Linux-based PCs, Apple PCs or even tablets.

Two subunits of the nuclear pore complex were immunolabelled using antibodies against gp210 and antibodies with multiple specificities (PAN4/5) and secondary antibodies coupled to Abberior STAR580 and Abberior STAR635P. Note that gp210 is localized in an eightfold symmetric structure at the rim of the nuclear pore complex. Shown is RAW DATA. Images were acquired using a STEDYCON attached to a Zeiss Axio Imager Z2.

  • Excitation laser lines: 450nm, 594nm, 640nm (all pulsed)
  • STED laser: 775nm (pulsed)
  • 2D STED resolution: <40nm, typically 30nm (depending on dyes and objectives used)

  • 3 confocal imaging channels
  • Detection: APD1: 650-700nm; APD2: dual-band detection: 502-572nm & 608-627nm, gated detection (for confocal and STED)
  • QUAD beam scanner: 90µm x 80µm (with a 100x objective); 512x512 pixel up to 1.1 frames/s; line frequency up to 800Hz
  • Pinhole: 1.1 Airy unit for a 100x objective
  • Software: browser-based control software operational on any modern PC, Mac or tablet: line interleaved STED and confocal imaging in three imaging channels; options to define z-stacks, acquisition queues and time series, auto-save function
  • Dimensions: STED-camera head: 11cm x 20cm x 20cm; controller: 22cm x 55cm x 60cm; total weight head plus controller unit < 40kg

The Abberior STEDYCON is compatible with any modern upright or inverted microscope. The following bodies have been experimentally tested:

  • Zeiss Axio Imager Z2 (Upright) with objective 100x NA=1.46
  • Nikon Eclipse Ni (Upright) with objective 100x N=1.45 Lambda Series
  • Olympus IX83 (Inverted)
